Blocking GTP hydrolysis during ribosomal translocation abolishes reverse rotation of the 30S head domain. We propose that – rather than driving tRNA movement – GTP hydrolysis is used as a checkpoint to ensure productive translocation and strict maintenance of the translational reading frame.
