Public Summary

We study aparted 3’UTR and cognate CDS expressions in hESCs and early embryos. We found that both in vitro and in vivo, hESCs and embryos self organizing into different structures defined by dynamic differential expressions of 3’UTR/CDS. We found that pluripotent genes shows unique expressions of 3’UTR apart from cognate CDS, and the cells highly expressing 3’UTR are also characterized by various histone methylation markers that indicate special condition or status of these cells. Further depletions of 3’UTR or histone methylation regulator gene proved that they are critical of both normal cell function and heterogeneous hESC structure formation.
